PHILADELPHIA (July 30, 2012) When the longest cells in the body are injured at their farthest reaches, coordinating the cells' repair is no easy task. This is in part because these peripheral nerve cells can be extremely long up to one meter in adult humans which is a lot of distance for a molecular distress signal to cover in order to reach the "command center" of the cell's nucleus.

Scientists have believed this process to be even more challenging because their textbook understanding for many years has been that the axons the long extensions of nerve cells away from the main cell body containing the nucleus do not manufacture the proteins involved in the molecular signal themselves. Yet, in recent years, some scientists have begun to challenge that textbook understanding, with preliminary evidence that one key protein involved in setting off a distress signal for cellular repair, known as importin beta1, was locally produced in the axons. They just weren't sure how.

"Now these textbooks need to be rewritten," said Dr. Jeffery Twiss, a professor and head of the department of biology in Drexel University's College of Arts and Sciences. Twiss co-authored new research recently published in Neuron, led by collaborators from the Weizmann Institute of Science. "Our new research is one of the strongest indicators yet of molecular signaling from end to end in peripheral nerve cells."

These researchers have provided strong new evidence that the protein importin beta1 is indeed produced locally in the axons of peripheral nerve cells. They also found that the version of the protein, when found in the axon, is made using a different molecular recipe than the version found in the nucleus, where it performs different essential cell functions. These discoveries may help scientists better understand how subsequent steps operate in the distress signal and in nerve cell repair, so they can eventually control and enhance the process to speed up recovery from nerve injuries.

Finding this evidence was far from simple: Importins are so crucial in the cell's nucleus that even the smallest embryo could not survive without them. But Rotem Ben-Tov Perry, a research student at the Weizmann Institute who was lead author of the new study, found a way to distinguish the importin beta1 in the cell body from that in the axon: The axonal protein was apparently made from a longer version of messenger RNA, the cell's working recipe for building a protein. To see if they could selectively affect just the axonal version of the protein, the Weizmann researchers worked with Drexel's Twiss to take advantage of high precision knock-out technology. Rather than knocking a whole gene out of the system, they managed to remove one little piece of the messenger RNA's recipe for manufacturing importins -just the longer bit that sends the RNA to the axon.

Now they observed plenty of importin beta1 in the cell body, but none in the axons. With the axonal segment of RNA knocked out of the recipe for importin beta1, a mouse embryo still had the importin it needed near the nucleus of its cells to grow and develop into a living animal but it took much longer to recover from peripheral nerve injury. Genes that are normally active in response to nerve damage were activated to a lesser degree. All of this suggests that the importin beta1 that normally helps inform the extended nerve cell about injury is, indeed, produced locally in the axon and that this protein found in the axon is a key part of the nerve repair signaling process.

"The data shows conclusively that importin beta1 protein is produced in axons, Rotem's work has validated the importins' crucial role in nerve repair," said Dr. Michael Fainzilber, senior author and professor at the Weizmann Institute.

Twiss said that next steps will be to better describe how the signaling process involving importin beta1 delivers a signal to begin nerve cell repair and, ultimately, develop strategies to better control these molecular steps of the repair mechanisms to improve nerve cell regeneration after injury.

Cuba: Raul Willing to Meet with the U.S. ? Miami International ...

The latest news coming out of Cuba is interesting, and funny, and all of those reactions typically engendered when someone says something peculiar.? Raul Castro, who by most accounts is still a puppet to his brother Fidel, is willing to meet with the U.S. so long as Cuba is not condescended to like a U.S. puppet, and is treated as an equal to the U.S.? Well, we?re broke, but we ain?t as broke as Cuba.? In fact, no one in the hemisphere is except for Haiti and perhaps Nicaragua.? Nothing to be obstinate about by any stretch of reason.? For its part, the U.S. said Cuba must improve rights and more before any meeting could occur. Message to Raul: All things such as this will have to wait until our elections are over in November, and you better pray for an Obama victory, ?cause Mitt will be beholden to the party line:? ?Just say ?no??to Cuba?.

Friends of Animals Utah (FOAU) : adoptashelter.com

Friends of Animals Utah (FOAU) is a private non-profit 501(c)(3) organization founded in 1990 by a group of animal lovers determined to find homes for animals in need. FOAU envisions a community where every pet has a permanent and loving home, every pet is spayed or neutered, and companion animals are treated with respect and compassion.

Since its founding, the organization has rehomed more than 10,000 needy pets. Over the decades, FOAU?s objective has remained the same: to place animals into permanent, caring homes and to advance the humane treatment of animals through education. Presently FOAU rescues over 1,100 animals per year, with about 100 pets in the system at any given time.

Each week, FOAU staff visits animal shelters throughout Northern Utah and rescues adoptable and treatable dogs and cats. Many of these animals are pulled from the shelters? euthanasia lists. These animals find themselves homeless for a variety of reasons ranging from abandonment to rescue from an abusive situation to owner surrender. Animals are taken to FOAU Rescue & Rehabilitation Ranch, the 16,000-square-foot facility where they receive medical care, are spayed/ neutered, microchipped and vaccinated. FOAU provides the same level of care consistent with what is given in a loving home. They?re also committed to assisting special needs animals. The facility (and other programs) are manned by four fulltime and ten part time employees in addition to an army of volunteers.

Once the animals are healthy, they go to the Furburbia adoption center located at Tanger Outlet Center in Park City, Utah. While cats can live at Furburbia, dogs must commute daily from the ranch to the adoption center. This gives a whole new meaning to the phrase, ?back at the ranch.?

The organization recently started the Purple Paw Project that cares for the pets of domestic violence victims. It is well-documented that many victims won?t leave abusive partners because they fear for the safety of the pets they must leave behind?and with good reason. A study of women seeking temporary ?safe haven? showed that 71% of those with pets reported that their partners had threatened, hurt, or killed their animals.

The Purple Paw Project provides a support service to domestic violence shelters which are unable to accept pets. It offers temporary shelter for the pets of domestic violence victims, allowing survivors and their children to seek shelter, medical treatment and counseling, knowing that their pets are safe.

FOAU houses the pet free of charge for the entire period that the client resides at the domestic violence shelter. The pets receive food, water, medication administration, daily socialization, and exercise. FOAU shelters the animals at a network of secure locations throughout the area. Initial capacity is estimated at 600 pets per year.

In April 2012 FOAU started its education outreach program that teaches school children about humane treatment of animals as well as the importance of spaying and neutering. This fall FOAU will also launch a no-cost spay/neuter and education program in Summit County.

I was thinking about being a babysitter for other people?s kids (obviously) but I was wondering if I?m watching several people?s kids at once regularly at what point would it become a home daycare? I?m just trying to figure out if I need to get a degree in early childhood, get lienced to be a daycare director or what. I just don?t want to get into this and all of a sudden I?m in trouble because I missed something some where. Thanks for your help!

VANCOUVER, British Columbia (AP) -The Vancouver Whitecaps could not stop David Beckham when they needed to Wednesday night.

Beckham scored a goal and assisted on another as the L.A. Galaxy roared back from a 2-0 deficit to tie the Whitecaps 2-2 before a disappointed sellout crowd of 21,000 at B.C. Place Stadium.

"Anytime you're 2-0 up and you tie 2-2, it feels like a defeat - that's obvious," Whitecaps coach Martin Rennie said. "But it was an exciting game, an entertaining game, a fantastic crowd tonight and a great atmosphere."

The defending-champion Galaxy (8-10-3) remain four points back of the fourth-place Whitecaps (8-6-7) in the Western Conference.

"We can't complain," L.A. coach Bruce Arena said. "We're getting out of town real quick here (Thursday) and not looking back at all. We're fortunate to get a point."

Second-half substitute Jose Villarreal, 18, secured the tie as he netted his first career MLS goal in the 87th minute. Beckham launched the play as he lobbed a lead ball to Juninho, who flicked it on to Villarreal.

The Inglewood, Calif., native, who was playing only his second game after missing most of the season following surgery on both shins, made no mistake as he turned and beat Vancouver goalkeeper Joe Cannon with a high shot.

"They obviously had a lot of play in the first half, and they played well," said Beckham. "But we were able to capitalize on the hard work that we put in towards the end of the game."

Beckham and Villarreal scored six minutes apart to cap the successful comeback.

Gershon Koffie and Barry Robson, with his first goal as a Whitecap and 100th of his professional career, scored for the Whitecaps in the first half. The tie spoiled a strong effort from Robson in his home debut after the Scottish international began his Vancouver tenure with four road games.

Robson, one of Vancouver's three designated players, sent four shots on goal while taking seven altogether.

"I think after a two-month holiday and coming straight and travelling to four different cities in two different countries I did all right," said Robson, who signed with the Whitecaps in February but had to complete his season with Middlesbrough in England first.

Robson and Beckham went toe-to-toe at times and argued with each other, especially in the late going. But all was forgiven afterward, said Beckham.

"Obviously, we're both passionate about the game and both passionate about winning," said Beckham. "He's Scottish, I'm English, so there's always a little bit of fight there. We went at it a little bit. We spoke after and were fine."

"He's a terrific footballer and a great ambassador for the game," said Robson. "A lot of people look up to him, and he's a nice guy, but on the pitch it doesn't matter who anybody is. At the end of the day I'm trying to win for my team and he's trying to win for his."

A minute after Vancouver's Koffie hit the crossbar, Beckham put the Galaxy on the scoreboard when he fired in a rebound from the top edge of the 18-yard box. The goal came after Bryan Gaul's curling shot hit the corner of the crossbar and post.

It was the third goal in two games for Beckham, but he did not take any consolation in playing the spoiler role in a draw after the crowd booed him the first time he touched the ball.

"I'd much prefer to play a winning goal," said Beckham. "But it's always nice to get a goal. I've scored a couple in the last few games, and it's been enjoyable, because I'm not used to it."

Koffie opened the scoring in the 17th minute following a turnover by Beckham. Vancouver's Brazilian striker Camilo blocked the Englishman's clearing attempt and the ball fell to Koffie. Camilo was knocked down as he ran into a defender, but play continued and Koffie put home a low left-footed shot off L.A. goalkeeper Josh Saunders' hand.

Robson also hit the corner of the post and crossbar from the right flank in the 84th minute. The midfielder was denied again twice in stoppage time as L.A. goalkeeper Saunders caught his shot and then knocked his free kick over the crossbar.

Arena credited Saunders with helping his squad escape with a draw.

"At the end of the game, he had to make some big saves - and he did," Arena said.

That's the conclusion of a new analysis published in ACS' journal Chemical Research in Toxicology.

Laboratory mice and rats serve as stand-ins for people for research that cannot be done on humans. In the article, Gale Carey and Lisa Merrill point out that the millions of lab rodents used in laboratory studies each year have a nutritional status that is different from other test animals. While other test animals are fed meals, rodents have round-the-clock access to food. And eat they do, gaining more weight and more body fat than meal-fed rodents. The authors cite other research indicating that lab rodents with free access to food tend to develop abnormally high blood fat levels, high cholesterol, nerve and heart damage, cancer and other disorders.

Their analysis of 54 studies concluded that having free access to food is likely to affect the results of tests for the toxicity and cancer-causing effects of new drugs and other substances in rodents, and could be the reason why such studies have been varying so much in recent years. "Therefore, it is crucial that feeding regimen be carefully considered in designing toxicology experiments," say the authors.

School Discipline Alternatives Explored In Harkin Hearing

During Elijah's early years in school, Debbie Jackson, his single mother, routinely came home to hear messages of his screams on her answering machine -- and no other word from administrators. The blood-curdling voicemails sometimes came as often as three times a day. "It was a nightmare," Jackson said. "I no longer recognized my son."

Elijah was screaming because he was an unruly kid, and his teachers tried to get him in line through seclusion and restraint. He would throw pencils on the floor, turn over desks, hit other students and rip pictures from the wall. Teachers did what most U.S. educators have been trained to do when faced with kids who act out violently. They restrained Elijah by his arms, wrists, and legs, or placed him in a locked room by himself.

Elijah got better after he transferred to a school that focused on positive behavior modification. That's why Jackson testified at Thursday's Senate Health, Education, Labor and Pensions committee hearing on seclusion and restraint in schools.

The hearing, the first time Congress has explored the issue, came after recent U.S. Education Department data shows schools physically restrained students 39,000 times during the 2009-2010 school year -- and black, Hispanic and students with disabilities bore the brunt of that. According to the Education Department's Civil Right Data Collection, a survey of 85 percent of the country's 100,000 schools released in March, 70 percent of students who were restrained had disabilities. Schools mechanically restrained African American and Hispanic students at twice the rate expected given their population.

The hearing also comes after several highly-publicized cases in which these tactics injured or killed students. ?In April, at the Leake and Watts School in Yonkers, New York, 16-year-old Corey Foster was restrained by school staff members who were trying to remove him from a basketball court," Harkin said. "Corey stopped breathing, went into cardiac arrest, and died." Corey's mother attended the hearing.

The people who testified at the hearing shared a common theme: While physical restraint seems like a logical reaction to kids who act out violently, the practice is damaging and costly, and doesn't usually fix behavioral problems. What works, the witnesses said, are preventative programs that replace disruptive behaviors with positive, benign patterns. And what needs to change in schools, they said, is training teachers to reinforce kids' good behavior.

Only 16 states have laws that limit the use of physical restraint in school, and the federal government has nothing on the books. Last year, Harkin sponsored a bill that would ban the use of seclusion and restraint in schools, barring extreme circumstances. Harkin had hoped to include it in the rewrite of No Child Left Behind, but that process is stalled.

A survey by the American Association of School Administrators released Wednesday said schools are cutting back on nonviolent intervention training. In 2009, the Safe and Drug Free Schools' state grant was eliminated, and the majority of AASA members reported the loss made it "considerably or moderately more difficult" to fund crisis training programs. the survey said 91 percent of the association's members reported that their schools would be better if provided with more funding for positive interventions.

But the association said it doubts that banning seclusion and restraint will work for all students, writing it is "unrealistic" to assume that positive behavior interventions by themselves will help special education students "as successfully as general education students." The group also said Harkin's bill is flawed because it rules out less harmful types of restraint and seclusion methods.

When Michael George took over Lehigh University's Centennial School, an alternative school in Pennsylvania that targets students with aggressive behavioral issues, he was disturbed, he said. "The use of physical restraint was commonplace," he told senators. In 20 days, staff members restrained students 112 times, often using a "basket hold" grip from behind. By year's end, George had closed the school's two seclusion rooms, and only used physical restraint once during the last 40 days of school. "We have the technical knowledge necessary ? to end the overreliance of seclusion and physical restraint," he said.

George said he changed his school by training teachers to solve problems with nonviolent behavior modification. The training helped Elijah, who is now 9. "I was not used to Elijah being treated as a person," Jackson, his mother, said. The school gave him a point system for good behavior that accrued into credits for shopping at the school score, a Burger King lunch, and field trips. "Centennial held Elijah responsible for his choices," Jackson recalled. Now, Elijah attends a traditional public school and wins awards for writing and art. He begins fourth grade in the fall.

You wanted to know: who are these scientists? Introducing: Ben Van Mooy (sort of)

So far we?ve met five of the six principal investigators of the cruise: Kay Bidle, Jack DiTullio, Marco Coolen, Anna Martins and Assaf Vardi. The only one left is Ben Van Mooy.

Broadly, Ben?s lab works on marine chemistry. They ask questions about things like how lipids are formed and when. Lipids are the main component of cell walls ? and there are lots of different types. The cell can change the composition of its cell membrane ? which lipids it?s building its little house out of ? based on the environmental conditions. If it?s hot, or cold, or salty, or the cells are stressed, they?ll swap out certain lipids in favor of others. Kind of like how we might change the types of curtains we use in our house to keep light or heat in or out depending on the season.

But I have to confess? I?ve never met Ben Van Mooy. I hear that he?s tall, and I know that he works at the Woods Hole Oceanographic Institution, but otherwise he is quite a mystery to me, since he?s not onboard.? He does have three representatives here, so we?ll meet them instead.

Justin thinks we should have worse weather.

Justin Ossolinski is a research technician in Ben?s lab. On board, Justin?s main job is to coordinate the team?s big instruments. The Van Mooy group has a bunch of instruments with them, and Justin he helps toss those instruments off the side of the boat and then drag back out. That includes big nets, traps and sensors.? These things are heavy, expensive and complicated. The ship has all sorts of expertise and special parts to help make sure instruments get into the water safely and come back out alive. Simply finding these traps again can be tricky, and watching the ship idle up just a few feet from the buoys bobbing along is pretty impressive.

This role, of being outside and hauling in heavy things, suits Justin quite well. It?s hard to imagine him sitting at a lab bench for too long, or, in fact, sitting anywhere for too long ? even to sleep. Which isn?t to say that he?s fidgety or easily excitable. Rather, if he?s not helping, hauling, or being useful somehow, he?s probably trying to find a way to be of assistance. That?s just how he is.

Bethanie Edwards also helps with the deck operations (deck-ops, as everyone calls it) alongside Justin. Bethanie is a second year PhD student in Ben?s lab.

Bethanie thinks the weather is great.

Along with deck-ops, she?s doing experiments on enzymes, lipids, and something called quorum sensing. The idea is that cells in the water can sense and signal to one another. By doing so, they can regulate how much, and by what processes they consume different nutrients. Sometimes the signals might be mutually beneficial ? triggering a collective action to consume nutrients in the water. Other times they might be selfish ? some microbes might try to trick others and keep all the nutrients to themselves. Bethanie looks at a couple different molecules that might be involved in quorum sensing, and when they seem to be more abundant in the water.

Bethanie is also studying for her qualifying exams at WHOI ? the test that determines whether she can continue forward in her PhD. Which means we?ve both spent some time searching for quiet places on the ship. Turns out, there really aren?t any ? although the science chart room is about as close at you can get for a desk without too much noise.

Also along with Ben?s group is Jamie Collins, a first year PhD student in Ben?s lab. Jamie?s got more experience at sea than most of the scientists on board after five years with the coast guard. So the Knorr feels roomy and luxurious to Jamie. He has kept his eye rolling to a minimum when I fret about 20 knot winds, and I appreciate that.

On board, Jamie?s in charge of an instrument called a pit trap ? a contraption that collects particles as they fall down through the water. The traps are made up of an array of tubes that are open at the top (a lot like the CTD) with tops that close at a designated time to trap the water. At the bottom of each tube is a mixture of super salty water meant to kill anything that falls down into the bottom. This keeps things from decaying and eating each other while the instrument is in the water.

Using the pit traps, Jamie can understand what kinds of organic matter is sinking down through the water column. It helps to tell him things about how carbon and phosphorous are produced and consumed in the ocean. The traps are also key to a lot of people on board ? from Marco?s work on marine snow to Daniella?s investigations of how the virus might carry carbon down through the water column.

The three of them correspond with Ben by email and phone calls, conveying the good and bad news and asking for advice. And all three seem to have a similar mentality about the cruise: making things work. There?s very little moaning and groaning when things break or don?t go the way they thought they would, but rather a quiet determination to fix it. When a few of their instruments were attacked by a sea bear-cat (or just roughed up in the surf, who knows) they got to work fixing them.

Later, I asked Bethanie what her favorite thing about being at sea is.? Immediately she said, ?It?s really challenging, and I kind of like that.? Basically, if I had to pick a team to travel with during some sort of apocalypse, I?d pick Ben?s.

You know the stereotypes already. Both Obamas come from what might loosely be called the intellectual/academic meritocracy, the "liberal elite," the post-WASP Ivy League, easily caricatured as the world of free-trade coffee, organic arugula, smug opinions, and Martha's Vineyard. The Romneys, by contrast, belong to the financial oligarchy, the "global elite," the post-financial-deregulation world which is just as easily caricatured as one of iced champagne, offshore bank accounts, dressage trainers, and private islands. The two groups have some important overlaps. Though Romney got some attention for holding a fundraiser in the Hamptons last week, it is Obama who has raised more money in the Hamptons overall (the president scored particularly well in Sagaponack, by one account, where the median home sells for $4.4 million).

Canadian scientists protest spending cuts

Several hundred Canadian scientists and their supporters held an unprecedented protest march on Tuesday to demonstrate against the government's decision to close down major facilities and fire research staff.

The protesters, who say the right-of-center Conservative government dislikes science, walked through central Ottawa behind a woman dressed as the Grim Reaper and a coffin designed to mourn the "Death of Evidence."

"Evidence is the way that adults navigate reality. To deny evidence is to live in a fairy world ... when countries engage in fantasy it's called state propaganda," Simon Fraser University professor Arne Moores told a crowd of around 800 people gathered on Parliament Hill.

The Conservatives intend to stop funding an Arctic environment atmospheric research laboratory, a move U.S. scientists complain will harm the world's ability to monitor the ozone layer.

Ottawa is also shutting down the Experimental Lakes Area in northern Ontario, which plays a major role in helping study the effects of water pollution. Budget cuts also mean hundreds of scientists and researchers will lose their jobs.

The government ? which will also kill off its own environmental advisory agency ? variously cites the need to save money and focus on more important areas.

"As a country we have been lagging behind our peer nations on applied research and commercialization and our government is taking steps to correct that ? but not at the expense of basic research," said a spokeswoman for Gary Goodyear, junior science minister.

Critics say Ottawa is more interested in helping the powerful energy industry and note the March budget included measures making it easier for major projects such as oil pipelines to receive building permission.

"The government is shutting down science, decimating Environment Canada ... it's pretty transparent," University of Waterloo earth sciences professor Sherry Schiff told Reuters.

Many in the crowd said it was the first time they could remember Canadian scientists demonstrating en masse.

At the end of the protest, some of those present placed flowers at the front of the coffin and science books inside it.

"An iron curtain is being drawn between science and society. Closed curtains, especially those made of iron, make for very dark rooms," said Jeff Hutchings, professor of biology at Dalhousie University in Halifax.

Not everyone sees the issue in this light. A post by a reader on the Canadian Broadcasting Corp's website dismissed the protest as "just more yapping from lefties who have had their year -round holiday collecting loon scat or buffalo farts canceled."
